目的:对两步离心法制备的雷公藤多苷自溶性微针中总二萜及总生物碱的含量进行测定。方法:采用紫外可见分光光度法,分别选择雷公藤甲素、雷公藤次碱为对照,雷公藤多苷样品经适当预处理后测定其中各有效部位的含量。结果:每片面积为2.75 cm2的雷公藤多苷自溶性微针中含总二萜2.17±0.11μg,总生物碱210.76±2.46μg。结论:首次将雷公藤多苷与微针剂型结合,雷公藤多苷自溶性微针中总生物碱的含量大于总二萜,微针中所含的总生物碱的含量为总二萜的100倍左右。
Abstract:Objective: The content of total diterpenoids and total alkaloid in dissolving microneedles of Tripterygium Glycosides prepared by two-step centrifugation method was determined.Methods: With triptolide and wilforine as the standards, total diterpenoids and total alkaloids were determined by UV-Vis spectrophotometry after suitable pretreatment of themicroneedles.Results: The area of each tablet is2.75 cm2 and the content of total diterpenoids was2.17±0.11 μg, the content of total alkaloids was210.76±2.46 μg.Conclusion: This is the first time that Tripterygium Glycosides have been combined with microneedle formulations, among two effective components, the content of total diterpenoids was the lowest, Followed by total total alkaloids.The content of total alkaloids in microneedles is about 100 times that of total diterpenoids.
